Platelet-derived growth factor (PDGF) exerts its stimulatory effects on cell growth and motility by binding to two related protein tyrosine kinase receptors, which can associate as three dimer forms (alpha/alpha, alpha/beta, beta/beta) with variable binding specificity for the PDGF ligands (1,2). The total number of the two receptor subunits and the ratio expressed varies between cell types and appears to account for the variable responsiveness of different cell types to PDGF (3). Ligand binding induces receptor dimerization and autophosphorylation, allowing binding and activation of cytoplasmic SH2 domain-containing signal transduction molecules including Grb2, Src, GAP, PI3 kinase,, PLCgamma and Nck. Applications: Suitable for use in ELISA, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilution: Immunohistochemistry (paraffin): 10-20ug/ml, ELISA: 0.5-1ug/ml, Western Blot: 1-2ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
