Anti-CD126 (Interleukin 6 Receptor, gp130, neutralizing, IL-6R)

The IL-6 receptor system consists of two chains, IL-6R alpha and gp130. The IL-6R alpha is the binding component speci?c to the IL-6, but does not transduce a signal alone. On the contrary, gp130 transmits signals not only of IL-6, but also IL-6 related cytokines, i.e. LIF, OSM, CNTF, IL-11 and CT-1. The low af?nity binding sites for IL-6, is composed of IL-6R alpha alone. At low concentrations, soluble IL-6R alpha acts as an agonist of IL-6 activity. CD126 is single chain type I transmembrane protein with Mr=130-140kD and is a common receptor subunit for many cytokines, forming complexes with CD126, IL-11R, LIF-R, and other proteins. These gp130-containing complexes mediate signaling of oncostatin M, IL-6, IL-11, LIF, CNF, and cardiolipotrophin 1. gp130 is expressed at low levels by most cell types. This antibody was recognized as reacting with gp130 at the Sixth Leukocyte Typing Workshop., , Applications: , Suitable for use in Neutralization and ELISA. Other applications not tested. Recommended Dilutions: ELISA (Capture): 2-8ug/ml , Neutralization: Measured by it's ability to neutralize IL-6 Ra induced inhibition of proliferation in the M1 mouse myeloid leukemia cell line. The ND50 is typically 0.02-0.1ug/ml in the presence of 30ng/ml recombinant human IL-6 Ra and 30ng/ml recombinant human IL-6., Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
