Endoglin is an endothelial homodimeric membrane antigen containing the tripeptide sequence RGD which is a recognition motif for adhesion receptors of the integrin family. CD105 is a Type I transmembrane protein and exists as an O-and N-glycosylated homodimer. Endoglin is a TGF beta-binding glycoprotein that is most abundantly expressed on endothelial cells in tissues with in?ammatory or in?ammatory-like characteristics, such as the skin lesions in psoriasis. Proliferating endothelial cells in granulation tissue and in tumors (e.g. melanoma of the skin) also exhibit enhanced expression of endoglin. In addition, a marked staining of syncytiotrophoblasts in placenta is observed. CD105 binds to TGF-b1 and b3 with high affinity but not TGF-b2. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry, Immunoprecipitation and Western Blot. Other applications have not been tested. Recommended Dilution: Immunohistochemistry: 1:10-1:50 Fresh frozen or acetone fixed sections. Not suitable for paraffin embedded sections., Flow Cytometry: < 1ug per 10e6 cells in 100ul., Western Blot: 1:500--1:1000. 90kD band observed under reducing condtions. Membrane preparations recommended, mouse vascular endothelial cells recommended as a positive control., Immunoprecipitation: Use rabbit anti-rat secondary antibody, followed by Protein A or anti-rat beads for recovery., Optimal dilutions to be determined by the researcher. Source: Tissue culture supernatant, Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
