Retinoblastoma protein, Rb protein, also known as pRB is a nuclear phosphoprotein that is encoded by the retinoblastoma tumor suppressor gene that regulates the G1/S transition of the cell cycle. Rb protein is phosphorylated by cyclin D1-Cdk4 and cyclin A/E-Cdk2 during the G1/S transition of the cell cycle. The acitivity of Rb protein is regulated by the phosphorylation and dephosphorylation of serine and theorine residues. Cyclin D1-cdk4 specifically phosphorylates Ser780 in Rb protein. The active form of Rb protein binds and activates transcription factors, including members of the E2F family, whose targets are necessary for S phase, A hyperphosphorylation form of Rb protein causes it to dissociate from E2F, leading to the induction of gene transcription, and progression into S phase. In a hyphophosphorylated form, Rb protein binds to a subset of E2F complexes and converts them to repressors who constrain expression of E2F target genes. Because of its central role in progression through G1, Rb protein is an important protein, involved in the integration of a number of signaling pathways that influence the cell cycle. It has been found that damage to the proteins that govern phosphorylation of Rb protein and therefore Positive Control exit of a cell from the G1 phase of the cell cycle are frequent in human cancers. It is therefore believed that inactivation of the ìRb pathwayî may be necessary for tumor development. Deletion or alteration of Rb, protein results in the childhood cancer Retinoblastoma (RB). RB is a congenital malignant tumor that arises from the nuclear layers of the retina., , Applications: , Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml (ECL), Optimal dilutions to be determined by researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
