Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a member of the histone H3 family. Transcripts from this gene lack polyA tails, instead, they contain a palindromic termination element. This gene is located separately from the other H3 genes that are in the histone gene cluster on chromosome 6p22-p21.3. Applications: Suitable for use in Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Immunocytochemistry: 2-4ug/ml. Positive immunostaining of chromatin in HeLa cells undergoing mitosis. Cells were ?xed with 4% paraformaldehyde and permeablized with 0.25% TritonX., Western Blot: 0.5-2ug/ml detected phospho-Histone H3 in colcemid treated HeLa cell lysate. , Optimal dilutions to be determined by researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
