Epidermal Growth Factor Receptor, also known as ErbB-1 (EGFR, a 185kD glycoprotein) is a transmembrane tyrosine kinase that regulates a variety of biological responses ranging from mitogenesis to stress signaling. The EGFR consists of a large extracellular domain, a single transmembrane domain and a cytoplasmic domain that exhibits kinase activity. Upon binding of EGF to the extracellular domain, the receptor undergoes dimerization and becomes phosphorylated on several tyrosine residues within the cytoplasmic domain. Autophosphorylation of tyrosine 1173 creates a major binding site for the protein tyrosine phosphatase SHP-1, which can dephosphorylate the EGFR and thereby block EGFR-induced activation of the ERK1&2 signaling pathway. Applications: Suitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml for AP/BCIP/NBT (MTT) detection, 0.1ug/ml for HRPO/ECL deterction, Use 0.5% Caseine, 0.5% BSA in PBST for blocking., Immunoprecipitation: 1-10ug per 10e6 vanadate treated A431 cells., ELISA: 0.05ug/ml, Optimal dilution to be determined by researcher. Positive Control: H2030-03H1-Cell lysate from vanadate-treated HepG2 cells., Add 100ul SDS-PAGE sample buffer and incubate at 90°C for 5 minutes. Aliquot and store at -20°C , Western Blot: 20ul/lane (mini gel) for HRP/ECL detection. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
