Chk1, one of the downstream protein kinases of ATM/ATR, plays an important role in DNA damage checkpoint, embryonic development and tumor suppression. The activation of Chk1 in response to blocking DNA replication and certain forms of genotoxic stress involves phosphorylation of Ser317 and Ser345. Chk1 is also phosphorlaed at Ser280 and Ser296 following DNA damage. Activated Chk1 can inactivate cdc25c via phosphorylation at serine 216, blocking the activation of cdc2 and transition into M-phase. Chk1 can also phosphorylate p53 at serine 20 in vitro. Applications: Suitable for use in ELISA, Western Blot, and Immunohistochemistry. Other applications not tested. Recommended Dilution: ELISA:1:4,000 , Western Blot:1:500-1:1,000, Immunohistochemistry:1:50-1:100, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
