Anti-phospho-4E Binding Protein-1 (Ser65) (4E-BP1, PHAS-1)

4E-BP1 (also known as PHAS-1) normally binds eIF4E, inhibiting cap-dependent translation. Hyperphosphorylation of 4E-BP1 disrupts this binding, activating cap-dependent translation. The PI3 kinase/Akt pathway and FRAP/mTOR kinase regulate 4E-BP1. 4E-BP1 is phosphorylated in vivo on multiple residues, phosphorylation by FRAP/mTOR on Thr37 and Thr46 of human 4E-BP1 may prime it for subsequent phosphorylation at sites including Ser65 and Thr70. Applications: Suitable for use in Western Blot, ELISA and Immunoprecipitation. Other applications have not been tested. Recommended Dilution: Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight., Immunoprecipitation: 1:50 , Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
