Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a member of the histone H3 family. Transcripts from this gene lack polyA tails, instead, they contain a palindromic termination element. This gene is located separately from the other H3 genes that are in the histone gene cluster on chromosome 6p22-p21.3. Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunocytyochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:2000-1:10,000 detects methylated Histone H3 in acid extracted proteins from HeLa cells. 1:2000-1:5000 dilution of a previous lot detected methylated Histone H3 in acid extracted proteins from HeLa and tetrahymena cells. HeLa cell acid precipitate was resolved by electrophoresis, transferred to nitrocellulose and probed with a 1:5000 dilution of H5110-14J. Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. , Chromatin Immunoprecipitation: 5ul immunoprecipitates methylated Histone H3 from1x10e6 3T3/A31 cells. , Immunocytochemistry: 1:500 shows positive immunostaining for methylated Histone H3 in L6 cells ?xed with 4% paraformaldehyde and permeabilized using 0.1% Triton X-100., Optimcal dilutions should be determined by the researcher. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
