Anti-dimethyl-Histone H3 (Lys4)

Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a member of the histone H3 family. Transcripts from this gene lack polyA tails, instead, they contain a palindromic termination element. This gene is located separately from the other H3 genes that are in the histone gene cluster on chromosome 6p22-p21.3. Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunocytyochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:2000-1:10,000 detects methylated Histone H3 in acid extracted proteins from HeLa cells. 1:2000-1:5000 dilution of a previous lot detected methylated Histone H3 in acid extracted proteins from HeLa and tetrahymena cells. HeLa cell acid precipitate was resolved by electrophoresis, transferred to nitrocellulose and probed with a 1:5000 dilution of H5110-14J. Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. , Chromatin Immunoprecipitation: 5ul immunoprecipitates methylated Histone H3 from1x10e6 3T3/A31 cells. , Immunocytochemistry: 1:500 shows positive immunostaining for methylated Histone H3 in L6 cells ?xed with 4% paraformaldehyde and permeabilized using 0.1% Triton X-100., Optimcal dilutions should be determined by the researcher.
