Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene contains introns and its mRNA is polyadenylated, unlike most histone genes. The protein encoded is a replication-independent member of the histone H3 family. Applications: Suitable for use in Peptide Inhibition and Western Blot. Other applications not tested. Recommended Dilution: Peptide Inhibition: 1uM of histone H3 peptide containing dimethyl-arginine 2 abolished, detection of histone H3 by anti-dimethyl-Histone H3 (Arg2) (1:1000 dilution) in immunoblot analysis of acid extracts from chicken erythrocyte cells. Acid extract proteins from, chicken erythrocytes were resolved by electrophoresis, transferred to nitrocellulose and, probed with anti-dimethyl-Histone H3 (Arg2) (1:1000, Lane 1), or with the antibody pre-absorbed with 1uM histone H3 peptides. Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Western Blot: 1:500-1:2000 detects dimethyl-histone H3 (Arg2) in acid-extracted proteins from chicken erythrocyte cells. No detection of unmethylated recombinant Histone H3 was observed. Recombinant Histone H3 and chicken erythrocyte cell lysate were resolved by electrophoresis, transferred to nitrocellulose and probed with antidimethyl-Histone H3 (Arg2) (1:1000 dilution). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. , Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
