Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. Acetylation of histone H3 occurs at several different lysine positions in the histone tail and is performed by a family of enzymes known as Histone Acetyl Transferases (HATs). Acetylation of lysine14 is commonly seen in genes that are being actively transcribed into RNA. Applications: Suitable for use in Western Blot, Immunocytochemistry and Chromatin Immunoprecipitation. Other applications not tested. Recommended Dilutions: Western Blot: 0.05ug/ml detected Histone H3 on 10ug of HeLa acid extract, sodium butyrate treated HeLa cells and recombinant Histone H3. Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemilumnescence detection system., Immunocytochemistry: 1:500 detected acetylated Histone H3 in NIH/3T3 and A431 cells., Chromatin Immunoprecipitation: 5ug using HeLa extracted chromain, Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb , I1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb I1904-46Q: IgG, H&L (HRP) Pab Gt xRb, Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
