Tumor Necrosis Factor-alpha (TNF-alpha) is a pleiotrophic pro-inflammatory cytokine secreted by various cells including adipocytes, activated monocytes, macrophages, B cells, T cells and fibroblasts. It belongs to the TNF superfamily of ligands and signals through two receptors, TNFR1 and TNFR2. TNF-alpha is cytotoxic to a wide variety of tumor cells and is an essential factor in mediating the immune response against bacterial infections. TNF-alpha also plays a role in the induction of septic shock, autoimmune diseases, rheumatoid arthritis, inflammation and diabetes. Applications: Suitable for use in Flow Cytometry. Other applications have not been tested. Recommended Dilutions: Flow Cytometry: 1:2. Use 10ul to label 5x10e5 cells in 50ul. Requires cell membrane permeabilization. , Optimal working dilutions to be determined by researcher. Hybridoma: Spleen cells from immunized BALB/c mice were fused with cells of the mouse X63 Ag8.653 myeloma cell line. Recommended Negative Controls: I1904-78R3: IgG1 Murine Negative Control (FITC), Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. , Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
