Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts. TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions. Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis. Applications: Suitable for use in ELISA, Flow Cytometry, Immunocytochemistry, Immunohistochemistry, Neutralization, Multiplex Detection and Western Blot. Recommended Dilution: ELISA: 1:2000-1:4000, Flow Cytometry: see references 6-12, Immunohistochemistry (frozen): see references 13-19, Immunohistochemistry (paraffin): see reference 16, Immunocytochemistry: see references 20 and 21, Western Blot: see reference 21, Neutralization: see references 1 and 22-24, Multiplex Detection: see reference 25, Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
