This gene encodes a protease inhibitor that regulates the tissue factor (TF)-dependent pathway of blood coagulation. The coagulation process initiates with the formation of a factor VIIa-TF complex, which proteolytically activates additional proteases (factors IX and X) and ultimately leads to the formation of a fibrin clot. The product of this gene inhibits the activated factor X and VIIa-TF proteases in an autoregulatory loop. The encoded protein is glycosylated and predominantly found in the vascular endothelium and plasma in both free forms and complexed with plasma lipoproteins. Several alternatively spliced transcript variants of this gene have been described, but the full-length nature of some of these variants has not been confirmed. Applications: Suitable for use in ELISA and Western Blot. Other applications have not been tested. , Recommended Dilutions: Peptide ELISA Titer: 1:64,000, Western Blot: 0.3-1ug/ml ~50kD band observed in lysates of cell lines HepG2 and HeLa (calculated MW of 35.0kD according to NP_006278.1). In transfected HEK293 transiently expressing TFPI bands of approx. 38kD to 45kD are observed. These bands are not observed in the non-transfected HEK293. , Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
