As a member of the serpin superfamily of serine protease inhibitors, SERPING1 is the physiological inhibitor of activated C1r and C1s, two serine proteases involved in the classical complement pathway. In addition, it inhibits plasma kallikrein and coagulation factor XIIa, two serine proteases involved in the processing of kininogen to release bradykinin. Therefore, it plays an important role in regulating activation of both the complement and contact systems. SERPING1 deficiency results in hereditary angioedema, which is characterized by recurrent episodes of localized angioedema of the skin, gastrointestinal mucosa or upper respiratory mucosa. The deduced amino acid sequence of human Serpin G1 precursor consists of 500 residues with a signal peptide. The mature protein of 478 amino acid residues is heavily glycosylated. Applications: Suitable for use in Direct ELISA, Western Blot and Immunoprecipitation. Other applications have not been tested. Recommended Dilutions: Western Blot: 1ug/ml using human lung tissue and human ovary tissue, Immunoprecipitation: 25ug/ml to immunoprecipitate from conditioned cell culture medium spiked with human SERPINGG1. , Optimal dilutions should be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
