Anti-Semaphorin 4D (BB18, A8, GR3, CD100 antigen, SEMA4D, CD100, SEMAJ)

Semaphorin 4D (Sema4D/CD100, previously sem J, G or C-like 2) is a widely expressed 150kD type I transmembrane glycoprotein of the Class 4 Semaphorin family with activity in the immune and nervous systems. The 861aa Semaphorin 4D contains a 710aa extracellular domain (ECD) with a sema domain and an immunoglobulin-like domain. Within the ECD region used as an immunogen, mouse Semaphorin 4D shares 87% and 93% aa identity with human and rat Semaphorin 4D, respectively. A soluble, active 120kD form is produced proteolytically upon activation of B and T cells. Semaphorin 4D binds Plexin B1, but also CD72 on B and dendritic cells. In developing neurons, Semaphorin 4D assists in guidance. Semaphorin 4D is proangiogenic when expressed by tumor-associated macrophages. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 2ug/ml, ELISA: 0.5-1ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS, sterile 40-50% glycerol, aliquot and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
