PTGIS (Prostacyclin synthase, also CYP8A1, PGIS and Prostaglandin I2 synthase) is a 5052kD (57kD predicted) microsomal, integral membrane member of Family 8 in the cytochrome P450 superfamily of molecules. It is expressed by skeletal and smooth muscle, fibroblasts, macrophages and endothelium. Prostaglandin I2/prostacyclin and thromboxane A2/TXA2 are two lipid mediators that are derived from the same precursor (PGH2). Acting upon PGH2, TXA2 is created through the action of TX synthase, while PGI2 is generated through the action of PTGIS/PGIS. Although they share a common precursor, they exhibit opposite effects on the vasculature, with PGI2 reported to block platelet aggregation, induce vasodilation, and inhibit smooth muscle proliferation. It apparently does so by binding to either the IP receptor or PPARgamma. Human PTGIS is 500 amino acids (aa) in length. It is a single span transmembrane protein that contains an Nterminal transmembrane segment (aa120) coupled to a lengthy 480aa cytoplasmic region. Over aa21500, human PTGIS shares 86% aa sequence identity with mouse PTGIS. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O or PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
