Prolactin (PRL) is a peptide hormone that exhibits high homology to growth hormone and is related to the peptide hormones of the interleukin family. The endocrine effects of PRL on human breast tissues include the regulation of growth and differentiation of ductal epithelium, proliferation and differentiation of lobular units, and initiation and maintenance of lactation. Prolactin-dependent signaling occurs as the result of ligand-induced dimerization of prolactin receptor (PRLr), a member of the cytokine receptor family. Heterogeneous expression of PRLr at the immunoblot and immuno- histochemical levels was observed in breast carcinoma specimens, with a statistically significant correlation between prolactin receptor and estrogen receptor expression. The quantitation of growth factor receptors such as estrogen and progesterone has been useful in the prognosis of human breast cancer. The presence of these receptors has also provided the basis for endocrine-based therapies, including tamoxifen. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: Direct ELISA: 0.5-1.0ug/ml. The detection limit is ~0.2ng/well., Western Blot: 0.1-0.2ug/ml. The detection limit is ~2ng/lane under non-reducing and reducing conditions., Immunohistochemistry: 5-15ug/ml. , Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
