PhyH is a peroxisomal protein that is involved in the alpha-oxidation of 3-methyl branched fatty acids. Specifically, this protein converts phytanoyl-CoA to 2-hydroxyphytanoyl-CoA. It requires iron and ascorbate as cofactors and is expressed in liver, kidney, and T-cells but not in spleen, brain, heart, lung and skeletal muscle. Defects in PhyH are a cause of Refsum disease (RD) and deficient protein activity has been associated with Zellweger syndrome and rhizomelic chondrodysplasia punctata. Alternate transcriptional splice variants, encoding different isoforms, have been characterized. Applications: Suitable for use in Western Blot, and ELISA. Other applications have not been tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile 40-50% glycerol, ddH2O. Reconstituted product is stable for 12 months at -20°C. Aliquot and store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
