Four distinct genes encode alkaline phosphatases in humans. The ALPP gene encodes the 67kD placental isozyme, also known as PLAP. In comparison, ALPI, ALPL, and ALPPL2 encode intestinal, liver/bone/kidney, and placental-like or germ cell APs, respectively. The placental alkaline phosphatase is a glycosyl phosphatidylinositol (GPI) anchored protein that is expressed on the cell surface as a homodimer. Human APs are ectopically expressed in several cancers, and their serum levels are useful tumor markers. Mature human Alkaline Phosphatase/ALPP shares 74% aa sequence homology with the mouse and rat enzymes. Applications: Suitable for use in Western Blot, Direct ELISA and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml using JAR human choriocarcinoma cell line, Immunoprecipitation: 25ug/ml using conditioned cell culture medium spiked with recombinant human Alkaline Phosphatase/ALPP, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
