Phosphoenolpyruvate carboxykinase (PEPCK) is a hormonally regulated enzyme responsible for the first committed step in gluconeogenesis, catalyzing the conversion of oxaloacetate to phosphoenolpyruvate. PEPCK mRNA is most abundant in the liver, kidney cortex and white adipose tissue, but is also found in brown fat, the mammary gland during lactation and the small intestine. Both non-insulin-dependent diabetes mellitus and streptozotocin-induced diabetes result in elevated PEPCK activity, protein and mRNA. Two isozymes of PEPCK have been identified, cytosolic and mitochondrial. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:200, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
