Product information "Anti-Paired Immunoglobulin-Like Type 2 Receptor alpha (PILRa, PILR alpha)"
Paired immunoglobulinlike type 2 receptor alpha (PILRa? also inhibitory receptor PILRalpha), are 55kD paired receptors that consist of highly related activating and inhibitory receptors, and are widely involved in the regulation of the immune system. PILRa is thought to act as a cellular signaling inhibitory receptor by recruiting cytoplasmic phosphatases like PTPN6/SHP1, and PTPN11/SHP2 via their SH2 domains that block signal transduction through of signaling, molecules. Human PILRa is synthesized as a 303aa precursor that contains a 19aa signal sequence, a 178aa extracellular domain (ECD), a 21aa transmembrane segment, and an 85aa cytoplasmic domain. The ECD contains one Ig like V type domain and one potential site for N-linked glycosylation The cytoplasmic domain contains two ITIM motifs (aa267272, and 296301). There are four isoforms for human PILRa A 38aa substitution is made for aa 264303 in isoform 2. Isoform 3 is missing aa152224 and there is a 24aa substitution for aa 152303 in isoform 4. Mature human PILRa is 45% aa identical to mature mouse PILRa PILRa, is predominantly detected in hemopoietic tissues and is expressed in monocytes, macrophages, and granulocytes, but not lymphocytes. It is also strongly expressed by dendritic cells. PILRa interacts with herpes simplex 1 glycoprotein B and functions as an entry coreceptor for this virus. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 2ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
