p63 is a transcription factor and member of the p53 gene family encoded by TP63 in humans with roles in epidermal development and lineage specification, as well as cell proliferation, death, senescence, and differentiation. Alternative promoter use and splicing of TP63 mRNA produces several variants, including alpha, beta, gamma, delta, and epsilon isoforms in full-length or N-terminally truncated forms known as TAp63 and DeltaNp63, respectively, which exhibit distinct functions, tissue expression, and cellular localization. Mutations in TP63 are associated with various developmental disorders, including ectrodactyly ectodermal dysplasia-clefting syndrome, split-hand/foot malformations, Rapp-Hodgkin syndrome, and primary ovarian insufficiency.
