NFATC1 (Nuclear factor of activated T cells c1, also NFAT2a) is a 120kD member of the NFAT family of transcription factors. NFATC1 is found in T cells and mast cells, and regulates cytokine transcription. NFATC1 is normally cytoplasmic and phosphorylated at Ser172. Upon a rise in intracellular Ca +2 , dephosphorylation occurs via calcineurin, and NFATC1 enters the nucleus. Human NFATC1 is 943aa in length. It contains a calcineurin-binding site (aa118-123), two transactivation domains (aa126-218 and 703-943), two NLSs (aa265-267 and 682-684), an NES (aa310-321) and an RHD that binds DNA (aa410-592). There are multiple isoforms. Individually or in common, there is an alternate start site at Met473, a deletion of aa826-943, a 29aa substitution for aa1-42, and a 19aa substitution for aa 698-943. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml using Jurkat human acute T cell leukemia cell line, Ramos human Burkitt's lymphoma cell line and Raji human Burkitt's lymphoma cell line, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
