Nitric oxide (NO) and reacti04.10.07ve oxygen species (ROS) are important mediators that produce harmful or protective functions. NO can react with superoxide anions (O2-), yielding the toxic oxidizing agent peroxynitrite (ONOO-). Peroxynitrite induces nitration of tyrosine residues leading to changes in protein structure and function, and alterations in signaling pathways. Antibodies to nitrotyrosine have been used to monitor the status of peroxynitrite-induced modification of proteins. Applications: Suitable for use in ELISA. Western Blot, though not tested, may potentially be used as an application. Other applications not tested. Recommended Dilution: Western Blot: 1-5ug/ml, ELISA: 1:10,000-1:50,000 using 50-100ng control nitrated proteins/well. , Optimal dilutions to be determined by the researcher. Western Blot Nitrated MW Standards: N2700-10 , Positive Control for Western Blot:, Proteins extracted from human A431 cells or other cells pretreated with peroxynitrite for 5 min, Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
