Neuropilin-1 is a type I transmembrane protein that is expressed in the developing nervous system and by endothelial and tumor cells. Neuropilin-1 binds members of the class III secreted Semaphorin subfamily as well as some isoforms of VEGF family proteins. The aa sequence of rat Neuropilin-1 extracellular domain is 98% and 93% identical to that of mouse and human Neuropilin-1, respectively. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ul labels 1-2.5x10e5 cells. Validated on bEND. cells and PC-12 cells., Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Cells may be Fc-blocked with 1ug of mouse IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction. , 2. After blocking, 10ul of N1276-91C was added to 1-2.5x10e5 cells and incubated for 30 minutes at RT., 3. Unbound antibody was removed by washing the cells , twice in Flow Cytometry Staining Buffer (Catalog #F4568-10). Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Mouse Lyse Buffer., 4. The cells were resuspended in Flow Cytometry Staining , Buffer for analysis by Flow Cytometry. As a control for this , analysis, cells in a separate tube should be treated with , CFS-labeled goat IgG antibody. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Carboxyfluorescein conjugates are sensitive to light.
