Nerve Growth Factor Receptor (NGFR) is important for the development, differentiation, and survival of variety of neuronal and non-neuronal cells. Its action is mediated by binding two distinct receptors, the high affinity p140 and low affinity Nerve Growth Factor (NGF) Receptor (p75NGFR) or Neurotrophin Receptor (p75NTR). Antibodies to NGF receptor may play a role in the identification of benign and malignant soft tissue lesions. Applications: Suitable for use in Western Blot, Immunohistochemistry and Immunocytocheistry/Immunofluorescence. Other applications not tested. Recommended Dilutions: Western Blot: 1:5000-1:10,000 using a whole cell extract of cultured NGF-differentiated PC12 cells., Immunohistochemistry (Frozen): 1:1000 on 4% paraformaldehyde perfusion-fixed, frozen, free-floating sections of rat brain, mid brain. Tissue post-fixation/embedding performed in 30% sucrose in PBS. Specifically stains cell bodies and dendrites., Indirect Immunofluorescence (IF-IC): 1:500 using NGF-differentiated PC12 cells. Fixation reagent #1 is 3-4% paraformaldehyde. Fixation reagent #2 is 0.5% Triton X-100. Specifically stains cell bodies., Optimal dilutions to be determined by the researcher. Positive Control: WB, IC/IF: PC12 cells treated with NGF (50ng/ml) for 4-5 days. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
