Metabotropic glutamate receptors have been classified into three groups. In general, class I receptors (mGluR1 and mGluR5) mediate the excitation of neurons, whereas activation of class II (mGluR2 and mGluR3) and class III (mGluR4a, mGluR7a, mGluR7b and mGluR8) results in a depression of synaptic transmission. Class I mGluRs are localized in the postsynaptic membrane and are positively coupled to phospholipase C (PLC). Class II and class III mGluRs localized to the presynaptic membrane and are negatively coupled to adenylate cyclase. It has been speculated that mGluR2 and/or mGluR3 may play a role in the modulation of the mossy terminal function. Applications: Suitable for use in Immunoblot, Immunoprecipitation, and Immunohistochemistry (paraffin embedded sections). Other applications have not been tested. Recommended Dilution: Immunoblot Analysis: 0.5-2ug/ml detects mGluR2/3 in 20ug of rat brain microsomal preparation. Note: Do not boil the microsomal preparation, heat to 37°C for 15-30 minutes. 20ug of rat brain microsomal preparation was resolved by electrophoresis, transferred to nitrocellulose and probed with M3884-76 (0.5ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system., Immunoprecipitation: 4ug immunoprecipitates mGluR2/3 from 400ug of rat brain microsomal preparation., Immunohistochemistry: Detects mGluR 2/3 in paraffin embedded rat brain tissue sections., Optimal dilution determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
