Microtubule-associated protein kinase kinase 1 (MEK1 or MAPKK1) is a member of a family of tyrosine/threonine protein kinases that activate the ERK1&2/MAPK enzymes by phosphorylating, both residues within the threonine - glutamic acid - tyrosine (TEY) motif in the activation loop. MEK1&2 are also activated by dual-phosphorylation, which occurs on serines 218 and 222, in the activation loop of the MEKs. Threonine 386 of MEK1 is phosphorylated by ERK2, which serves as a negative feedback loop by suppressing activation of MEK2. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 , Optimal dilutions to be determined by the researcher. Positive Control: NIH3T3 cells +/- PDGF, A431 +/- EGF, PC12 +/- NGF. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
