Mcl-1 is a member of the Bcl-2 family whose members fall into two groups with opposing functions in apoptosis. Pro-survival family members such as Bcl-2, Bcl-xL and Mcl-1 inhibit apoptosis whereas death-inducing members such as Bax, Bid and Bad have the opposite effect. Mcl-1 is an immediate-early gene activated by the GM-CSF as well as IL-3 signaling pathways and plays an important role in the viability response of these cytokines. The downstream half of the Mcl-1 encoded gene product contains BH1, BH2 and BH3 domains that characterize it as a member of the Bcl-2 family protein and Mcl-1 terminates in a TM domain. However, unlike other members of this protein family, Mcl-1 has an extended N-terminal domain rich in PEST sequences that is probably responsible for the short half-life of this protein (1). Recent data describes a splice variant of Mcl-1 known as Mcl-1s/ ?TM which is derived from exon skipping. The full-length human Mcl-1 product consists of three coding exons whereas the Mcl-1s/ ?TM variant represented the joining of the first and third exons. This does not affect the BH3 domain in the first exon but does affect the BH1, BH2 and TM domains downstream because of the skipped second exon and a change in the reading frame downstream. The features of the Mcl-1s/ ?TM encoded gene product are reminiscent of BH3 only pro-apoptotic family members such as Bid and Bad which promote cell death.
