Anti-Interleukin 6 Receptor (CD126, IL-6R)

Interleukin 6 Receptor (IL-6 R), is a transmembrane protein that associates with the signal- transducing subunit gp130 to form a high affinity receptor complex for IL-6. It is expressed on T cells, activated B cells, and peripheral monocytes. A soluble form of IL-6 R can be released by proteolytic cleavage. Within the extracellular domain, human IL-6 R shares 52% aa sequence identity with mouse and rat IL-6 R. Applications: Suitable for use in Neutralization, Western Blot, ELISA and Flow Cytometry. Other applications have not been tested. Recommended Dilutions: Neutralizationof Human IL6 R Bioactivity: ~0.04-0.2ng/ml will neutralize 50% of the bioactivity due to 20ng/ml or recombinant human IL-6R and 20ng/ml of recombinant human IL-6. Recombinat human IL-6R enhances recombinant human IL-6-induced growth inhibition of M1 cells in a dose-dependent manner. The ED50 for this effect is typically 5-15ng/ml in the the presence of 20ng/mk of recombinant human IL-6., Western Blot: 1-2ug/ml, The detection limit for recombinant human IL-6R is ~100ng/lane under non-reducing and reducing conditions. , ELISA Capture: 2-8ug/ml. Use biotinylated human IL-6 R detection antibody and recombinant human IL-6 R as the standard., Flow Cytometry: 25ug/ml, 10ul labels 1-2.5x10e5 U937 cells in a total reaction volume <200ul. , Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
