IL-3 is a hematopoietic growth factor that promotes the survival, differentiation and proliferation of committed progenitor cells of the megakaryocyte, granulocyte-macrophage, erythroid, eosinophil, basophil and mast cell lineages. Produced by T cells, mast cells and eosinophils, IL-3 enhances thrombopoieses, phagocytosis, and antibody-mediated cellular cytotoxicity. Its ability to activate monocytes suggests that IL-3 may have additional immunoregulatory roles. Many of the IL-3 activities depend upon co-stimulation with other cytokines. IL-3 is species-specific, variably glycosylated cytokine, Applications: Suitable for use in ELISA, Immunofluorescence and Flow Cytometry. Other applications not tested. Recommended Dilutions: ELISA Detection: Monoclonal antibody is the preferred detection antibody in a sandwich ELISA for quantifying human IL-3 protein levels. Biotinylated antibody should be paired with purified antibody as the capture antibody, with purified recombinant human IL-3 as the standard. Immunofluorescence/Flow Cytometry: This antibody is useful for intracytoplasmic staining and flow cytometric analysis to identify and enumerate IL-3-positive cells within mixed cell populations. Recombinant human IL-3, unlabeled antibody, or Biotinylated Rat IgG1 should be used as a specificity control. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
