Interleukin 23 (IL23) is a heterodimeric cytokine composed of two disulfidelinked subunits, a p19 subunit that is unique to IL23, and a p40 subunit that is shared with IL12 (1 5). The functional IL23 receptor complex consists of two receptor subunits, the IL12 receptor beta 1 subunit (IL12Rb1) and the IL23 specific receptor subunit (IL23R) (3). Human IL23 R cDNA encodes a 629 aa type I transmembrane protein with a 23 aa residue signal peptide, a 330 aa residue extracellular domain, a 23 aa residue transmembrane domain and a 253 aa residue cytoplasmic region. IL23 R shares structural features with the IL12 Rb2, including an Nterminal Iglike domain, two cytokine receptor domains and multiple glycosylation sites in the extracellular domain. IL23 R lacks the three extracellular membraneproximal fibronectintype III domains present on IL12 RB2. IL23 R has a WQPWS sequence in the transmembraneproximal cytokine receptor domain similar to the cytokine receptor signature WSXWS motif. The cytoplasmic region of IL23 R has three potential Src homology 2 domainbinding sites and two potential Statbinding, sites. The gene for human IL23 R is located on human chromosome 1 within 150kb of IL12, Rb2. Human and mouse IL23 R share 66% amino acid sequence identity. Based on quantitative realtime PCR, human IL23 R mRNA is expressed in a human Th1 and Th0 clone as well as several NK cell lines and clones. Low but detectable levels of IL23 R mRNA is also expressed in EBVtransformed B cells and activated PBMC. IL23 initiates a signal transduction cascade similar to that of IL12, and involves Jak2, Tyk2, Stat1, Stat3, Stat4, and Stat5. IL23 has biological activities that are similar to, but distinct from IL12. Conjugate: Excitation Wavelength: 488nm, Emission Wavelength: 515-545nm, Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: Neat. 10ul labels 1x10e6 cells., Optimal dilutions to be determined by the researcher. Storage and Stability: Store at 4°C. Do not freeze. Stable for 12 months at 4°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Protect from light, Carboxyfluorescein conjugates are sensitive to light.
