Interleukin 22 (IL-22), also called IL-10-related T cell-derived inducible factor or IL-TIF, is a secreted cytokine that contributes to the inflammatory response in vivo. IL-22 is expressed in T cells and mast cells in response to IL-9 or lipopolysaccharide. IL-22 monomers bind a class II cytokine receptor that is a heterodimer of CRF2-9 (IL-22 Ra1) and CRF2-4 (IL-10 Rb). Mouse IL-22 is a 179 aa protein with a 33 aa signal peptide, three N-glycosylation sites and two intramolecular disulfide bonds. The mature protein shows 78% and 90% aa identity with human and rat IL-22, respectively. Applications: Suitable for use in Western Blot, Flow Cytometry and Direct ELISA. Other applications have not been tested. Recommended Dilutions: Western Blot: 1-2ug/ml. Detection limit for recombinant mouse IL-22 is ~1ng/lane under non-reducing and reducing conditions. , Flow Cytometry: 25ug/ml. Add 10ul of the diluted solution to 1-5 x10e5 cells in a total reaction mixture not exceeding 200ul. For intracellular staining with PMA/ionomycin- stimulated EL-4 cells to detect IL-22, cells must first be fixed and permeabilized using 4% paraformaldehyde and 0.1% saponin in PBS. Secondary antibody such as anti-rat IgG conjugated to a fluorochrome can be used. , Direct ELISA: 0.5-1ug/ml. Detection limit for recombinant mouse IL-22 is ~2.5ng/well. , Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
