Interleukin 17 (cytotoxic T-lymphocyte-associated serine esterase 8) is a proinflammatory cytokine produced by activated T cells. This cytokine regulates the activities of NF-kappaB and mitogen-activated protein kinases. This cytokine can stimulate the expression of IL6 and cyclooxygenase-2 (PTGS2/COX-2), as well as enhance the production of nitric oxide (NO). High levels of this cytokine are associated with several chronic inflammatory diseases including rheumatoid arthritis, psoriasis and multiple sclerosis. Interleukin-17 (IL-17) and its cognate receptor, IL- 17R, are examples of cytokine receptor pairs. IL-17 protein exhibits a striking degree of homology with the HSV13 protein which mimics its function. Applications: Suitable for use in Neutralization, Western Blot, Immunocytochemistry, Flow Cytometry, Direct ELISA and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Neutralization: The ND50 is 0.02-0.12ug/ml in the presence of 15ng/ml of rhIL-17, using theNIIH/3T3 mouse embryonic fiboblasts, Immunocytochemistry: 5-15ug/ml using immersion fixed human peripheral blood mononuclear cells treated with PHA, Western Blot: 1ug/ml using human primary naive CD4+ T cells and human primary differentiated Th17 cells, Flow Cytometry: 2.5ug to label 1x10e6 cells using human peripheral blood mononuclear cells treated with PHA and Ca2+ ionomycin, fixed with paraformaldehyde and permeabilized with saponin, Immunoprecipitation: 3ug/100ug cell lysate using human primary differentiated Th17 cells, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
