Interleukin 1 alpha (IL-1 alpha) is synthesized as a 33kD precursor that mainly remains intracellular. Cleavage by calpain in keratinocytes, epithelial cells and endotoxin-stimulated peripheral blood mononuclear cells produces a 112aa, 16kD propeptide and a 159aa, 17kD mature protein. A nuclear localization site in the propeptide allows transit to the nucleus, where either the precursor or the propeptide can activate transcription or alter RNA splicing. This can increase motility or senescence of normal cells or increase apoptosis of tumor cells. Human and mouse IL-1 alpha propeptide share 71% aa identity. Applications: Suitable for use in Western Blot and Direct ELISA. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
