Rat IL-1 alpha is a 17.7kD cytokine which is produced by macrophages. IL-1 alpha is chemotactic for leukocytes and also contributes to the inflammatory response by its interactions with several other cell types. Interleukin 1alpha (IL1 alpha) and interleukin1 beta (IL1 beta) bind to the same cell surface receptor as IL-1 and elicit nearly identical biological responses, sharing 25% homology in their amino acid sequence. IL-1 alpha is a member of the interleukin 1 cytokine family. This cytokine is a pleiotropic cytokine involved in various immune responses, inflammatory processes, and hematopoiesis. This cytokine is produced by monocytes and macrophages as a proprotein, which is proteolytically processed and released in response to cell injury, and thus induces apoptosis. The gene encoding this protein and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2. It has been suggested that the polymorphism of these genes is associated with rheumatoid arthritis and Alzheimer's disease. Both IL-1alpha and IL-1beta binds to the same receptor and has similar if not identical biological properties. These cytokines have a broad range of activities including, stimulation of thymocyte proliferation, by inducing IL-2 release, B-cell maturation and proliferation, mitogenic FGF-like activity and the ability to stimulate the release of prostaglandin and collagenase from synovial cells. However, whereas IL-1beta is a secreted cytokine, IL-1alpha is predominantly a cell-associated cytokine. Applications:, Suitable for use in ELISA, Western Blot. Other applications not tested. Recommended Dilutions: ELISA: 4ug/ml using 100ul/well to detect a range of 4-250pg/ml of recombinant rat IL1 alpha. Capture antibody in rat IL-1a sandwich ELISA, Western Blot: 50ng/lane under non-reducing and reducing conditions. Predicted molecular weight: 31kD. , Optimal working dilutions to be determined by researcher. Recommended Secondary Reagents: Donkey Anti Sheep/Goat IgG: HRP conjugate , Storage and Stability:, Lyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
