Integrins are a family of dimeric proteins that mediate cell-cell and extracellular matrix adhesion. Each VLA subtype (VLA-1, -2, -3, -4 and -5) is composed of a distinct alpha subunit noncovalently associated with a common beta subunit, as an alpha-beta dimer. Integrin VLA-3 (very late antigen, alpha 3/beta 1) is a cell surface receptor that binds to fibronectin, lamin, collagen type I and VI. VLA-3 is typically restricted in normal tissue but is widely expressed in many cultured tumor cells and correlates with the degree of invasiveness of malignant melanoma cells. Applications: Suitable for use in Immunohistochemistry, ELISA, RIA, Immunoprecipitation, Immunocytochemistry and FACS. Other applications have not been tested. Recommended Dilution: Immunohistochemistry: Acetone fixed, paraffin embedded sections, Immunoprecipitation: Under non-reducing conditions, two chains with apparent molecular weight of 110 and 150kD. In the presence of a reducing agent (2% B-ME), a single molecular species of 130kD is detected. Under reducing conditions both alpha3 and beta1 subunits run with the same apparent molecular weight. , Optimal dilutions to be determined by the researcher. Positive Controls: Cell Line: KJ29 renal carcinoma adherent cells, Tissue: Kidney. Reacts with basal layer podocytes and basement membrane of distal tubules. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
