TCCR, also known as WSX-1, is a class I cytokine receptor family member that is highly expressed in lymphoid tissues. It shows homology to IL-12 Rb2 and mediates IL-27 activity that is required for the initiation of Th1 responses. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 0.25ug-1ug/10e6 cells using mouse splenocytes., Optimal dilutions to be determined by the researcher. Hybridoma: Fusion of a mouse myeloma with B cells obtained from a rat immunized with purified, NS0-derived, recombinant mouse T Cell Cytokine Receptor. Flow Cytometry: 1. Cells may be Fc-blocked with 1ug of mouse IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction. , 2. After blocking, 10ul of I8443-81 was added to 1-2.5x10e5 cells and incubated for 30 minutes at RT. , 3. Unbound antibody was removed by washing the cells twice in Flow Cytometry Staining Buffer. Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Mouse Lyse Buffer. , 4. The cells were resuspended in Flow Cytometry Staining Buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with CFS-labeled rat IgG2B antibody. This procedure may need to be modified, depending upon the cell type and final utilization. Individual users may need to titrate to determine the optimal reagent amount for their specific use. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Caution: Carboxyfluorescein conjugates are sensitive to light.. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Fluorescein conjugates are sensitive to light.
