Heat shock proteins, HSP's or stress response proteins SRP's, are synthesized in a variety of environmental and patholphysiological stressful conditions. Many HSP's are involved in processes such as protein denaturation-renaturation, folding-unfolding, transport-translocation, activation-inactivation, and secretion. HSP70 is found to be associated with steroid receptors, actin, p53, polyoma T antigen, nucleotides, and other unknown proteins. Also, HSP70 has been shown to be involved in protective roles against thermal stress, cytotoxic drugs, and other damaging conditions. Applications: Suitable for use in Flow Cytometry, Immunofluorescence, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilution: Immunoprecipitation (Native & denatured): 2ug antibody/mg protein lysate using Protein A., Western Blot: 0.5-1ug/ml for 2hrs at RT., Immunohistochemistry (Frozen & formalin/paraffin): 2-4ug/ml. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 min., followed by cooling at RT for 20 min., Optimal dilutions to be determined by the researcher. Positive Control: BT474 or HeLa cells, breast carcinoma. , Cellular Localization: Nuclear and cyotplasmic, Molecular Weight of Antigen: Inducible: 72kD, Constitutive: 73Kd, Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
