High mobility group protein B3 is a 23kD nuclear protein and member of the HMGB family. Human HMGB3 is synthesized as a 200aa precursor, which is demethylated to produce the 199aa mature chain. The protein contains two HMG box DNA-binding domains aa9-79-93-161 and an acidic Asp/Glu-rich region aa181-200. Human HMGB3 shares 98aa sequence identity with mouse and bovine HMGB3. HMGB3 is expressed predominantly in the placenta. It binds preferentially single-stranded DNA and unwinds double stranded DNA. Applications: Suitable for use in Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 2ug/ml, Immunocytochemistry: 8-25ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
