Anti-EGF-Receptor (Epidermal Growth Factor Receptor, erbB, EGFR)

The EGFr which tranduces thebiological effect of EGF is a 170kD glycoprotein containing two functional domains linked by a transmembrane region. The extracellular domain is glyosylated possessing a single binding site. The intracellular domain contains a protein tyrosine kinase. Binding of EGF to receptor stimulates tyrosine kinase activity, essential for propagation of signal transduction. Upon ligand binding, EGFr becomes autophosphorylated and also phosphorylates several other endogenous proteins including phospholipase cg. These events initiate a number of intracellular responses which include increased levels of intracellular Ca2+ and transient expression of the nuclear oncogene products c-myc and c-fos. Application(s): Western Blot: 1:50 -1:200. Dependent on receptor dilutions in the preparation., Immunoprecipitation: A-431 epidermiod carcionoma (EGFr = 2 million/cell). Range: 10-12ug/ml. Protein kinase activity is measurable on the immunoprecipitated receptor. , Immunocytochemistry 1:100-1:200. Formalin fixed cells. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
