Functions of the cell cortex, including motility, adhesion, and cytokinesis, are mediated by the reorganization of the actin cytoskeleton. Recent evidence suggests a role for the Formin homology (FH) proteins in these processes. DAAM1 contains FH domains and belongs to a novel FH protein subfamily implicated in cell polarity. Wnt/Fz signaling activates the small GTPase Rho, a key regulator of cytoskeleton architecture, to control cell polarity and movement during development. Activation requires Dvl-Rho complex formation, an assembly mediated by DAAM1, which is thought to function as a scaffolding protein. Applications: Suitable for use in FLISA, Western Blot, and Immunohistochemistry. Other applications not tested. Recommended Dilution: FLISA: 1:1,000, Western Blot: 1:50-1:100, Immunohistochemistry: 1:10-1:50, Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer.
