Cyclin D1 is a nuclear protein (36kD ). D type cyclins are predominantly expressed in the G1 phase of the cell cycle. Cyclin D1 study is of interest in various forms of malignancy and over expression has been linked to poor prognosis. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry (frozen and paraffin), ELISA, Immunoprecipitation and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:2000, Flow Cytometry: Neat, Use 5ul of the optimal working dilution to stain 10e6 cells in 100ul. Permeabilization is recommended prior to staining., Immunohistochemistry (Paraffin and frozen): 1:1000-1:10,000 Note: Paraffin sections require antigen retrieval using sodium citrate buffer, pH 6.0, heat treatment prior to staining., Optimal working dilutions to be determined by researcher. Recommended Positive Control Tissue: Small intestine, colon. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
