G1:S-specific Cyclin-D1 protein is encoded by the CCND1 gene and it belongs to the highly conserved Cyclin Family. Cyclin family members, including G1/S-specific Cyclin-D1, are characterized by a dramatic periodicity in protein abundance throughout the cell cycle. Cyclin-D1 acts as an important regulator of progression from G1 to S phase. Degradation of Cyclin-D1 results in arrest in G1 phase. Accumulation of Cyclin-D1 is regulated at multiple levels including transcription, post-translational activation and cellular localization throughout the cell cycle. Cyclin-D1 also acts to promote cell migration by inhibiting Rho/ROCK signaling and expression of thrombospondin-1 (TSP-1), an extracellular matrix protein that regulates cell migration in many settings including cancer. Mutations, amplification and overexpression of this gene, which alters cell cycle progression, are observed frequently in a variety of tumors and may contribute to tumorigenesis in humans. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 4-6ug/ml, Immunohistochemistry (formalin fixed paraffin embedded): 5ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
