Anti-CD274 (B7-h1 Protein, B7-H1, B7H1, MGC142294, MGC142296, PD-L1, PDCD1L1, PDCD1LG1, PDL1)

CD274 is expressed constitutively on macrophages and dendritic cells, and is induced on activated T-cells, B-cells, endothelial cells and epithelial cells in response to Interferons alpha, beta and gamma. CD274 is reported to possess dual functions, inhibition of activated effector T cells and co-stimulation of naïve T cells. CD274 inhibits proliferation of activated T cells via ligation to the co-inhibitory molecule CD279 (programmed death-1, PD-1) leading to the secretion of the regulatory cytokine interleukin-10. CD274 has also been shown to co-stimulate early T cell priming and differentiation. Deregulated CD274 function has been reported in chronic viral and intracellular bacterial infection, as well as in many autoimmune diseases and cancers. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 0.5-1ug/ml. Detects a band of ~32kD in Raji cell lysate., Immunohistochemistry (Paraffin): 2.5ug/ml. Requires antigen retrieval using heat treatment prior to staining paraffin sections. Sodium citrate pH 6.0 is recommended., Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1903-30U: IgG, H&L, X-adsorbed (HRP) ShxRb, Recommended IHC Positive Control Tissue: Human heart, Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
