PVR is a member of Ig-superfamily with 3 Ig-domains in arrangement V-C-C. Four mRNAs are produced from single gene for PVR, (mapped to human chromosome band 19q13.1-13.2). Two of the mRNAs (called H20A and H20B or alpha and kappa,) encode cell surface molecules of about 43kD and 45kD polypeptide backbone, respectively. Two other splice variants, Beta and gamma encode secreted molecules of about 44kD. The cytoplasmic tails are short (aa 35-50) and are rich in serine phosphorylated residues. C2548-85 protects HeLa cells against the cytopathic effect of all three poliovirus serotypes. Intact Ig or Fab fragment of C2548-85 prevents binding of S-labeled poliovirus to HeLa cells. Applications: Suitable for use in Flow Cytometry and Immunofluorescence, Inhibition of Replications of Polio virus in HeLa cells use C2548-85X. Not suitable for use in Western Blot. Other applications have not been tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Positive Control: HeLa, SV80, HE-p2, RD, A431, HL-60, U-937 cells, monkey vero, GMK and TC-7 cells. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
