CCR3 is a G protein-linked seven transmembrane chemokine receptor that preferentially binds eotaxin and eotaxin-2. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ul labels 1x10e6 human peripheral blood neutrophils and CCR3-transfected Y3 cells, Absorption: 482 and 564nm, Emission: 675nm, Optimal dilutions to be determined by the researcher. Hybridoma: Spleen cells from immunized rats were fused with cells of the Y3 myeloma cell line. Flow Cytometry Protocol: 1. Fc-block cells with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction., 2. After blocking, add 10ul of C2099-92Q to 1x10e6 cells and incubated for 30 minutes at RT., 3. Wash the cells twice in F4568-10: Flow Cytometry Staining Buffer to remove unbound antibody. Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Human Lyse Buffer., 4. Resuspend the cells in F4568-10: Flow Cytometry Staining Buffer for final Flow Cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with Percp-labeled rat IgG2A antibody. This procedure may need to be modified, depending upon the cell type and final utilization. Individual users may need to titrate to determine the optimal reagent amount for their specific use. Storage and Stability: May be stored at 4°C. For long-term storage, aliquot and store at 4°C. Do not freeze. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer. PerCP conjugates are sensitive to light.
