The BH3 domain-only protein BID, a death agonist member of the Bcl-2/Bcl-xL family, is localized in the cytosolic fraction of cells as an inactive precursor. Its active form is generated upon proteolytic cleavage by caspase-8 in the Fas signaling pathway. Cleaved BID translocates to mitochondria and induces cytochrome c release and mitochondrial damage. Thus, BID relays an apoptotic signal from the cell surface to mitochondria. However, the precise molecular mechanism for the translocation of the cleaved BID and for the subsequent release of cytochrome c during apoptosis is unclear. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1:1000. Incubate membrane with diluted antibody in TBS, 5% BSA, 0.1% Tween-20 at 4°C with gentle shaking, overnight., Immunoprecipitation: 1:200, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
