Product information "Anti-Bcl-XL"
Bcl-xL (28kD) is part of the Bcl-2 family, which are important regulators of apoptosis. Bcl-X has two isoforms, Bcl-xL (long) a 241 amino acid protein and shares signi?cant homology with Bcl-2. Bcl-xS (short) a 178 amino acid protein lacking a 63 amino acid domain near the transmembrane region that is well conserved among members of the Bcl-2 family. Like Bcl-2, Bcl-xL has the ability to inhibit apoptosis and is widely studied for its role in tumor generation. In a recent study, analysis of Hodgkin's disease showed that 71% of cases (114/160) showed immunohistochemical reactivity with Bcl-xL in Reed-Sternberg cells. In some cases, Bcl-xL reactivity was present in Reed-Sternberg cells that were negative for CD15 and CD30. Atypical hyperplasias (0/28) were negative for Bcl-xL., Bcl-xL prevents apoptosis through two different mechanisms: by heterodimerization with an apoptotic protein to inhibit its apoptotic effect (1,2), and by its direct pore-forming effect on the outermembrane of mitochondria to help maintain a normal membrane state under stress conditions (3). Applications: Suitable for use in ELISA, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:1000, Immunoprecipitation: 1:50, Immunohistochemistry (paraffin): 1:25. Antigen retrieval by heating in citrate buffer is required., Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
